Regenerative Effect of Mesenchymal Stem Cell on Cartilage Damage in a Porcine Model

Osteoarthritis (OA) is a major public and animal health challenge with significant economic consequences. Cartilage degradation plays critical role in the initiation progression of degenerative joint diseases, such as OA. Mesenchymal stem cells (MSCs) have become increasingly popular field cartilage regeneration due to their promising results. The objective this preclinical study was evaluate regenerative effects mesenchymal repair knee defects using porcine model. Seven healthy LYD breed white pigs, aged 9–10 weeks weighing approximately 20 ± 3 kg, were used experimental protocol. Full-thickness measuring 8 mm diameter 5 depth induced lateral femoral condyle posterior limbs both joints sterile puncture technique while maximally flexed. Following 1-week induction phase, pig treatment groups received 0.3 million/kg MSC transplant into damaged region, placebo group control solution treatment. Magnetic resonance imaging (MRI), computerized tomography (CT), visual macroscopic examination, histological analysis, cytokine concentration analysis assess regeneration. findings revealed that human adipose-derived (hADSCs) more effective repairing than umbilical cord-derived (pUCMSCs).
